Our strategic partnership is designed to combine the strengths and expertise of both companies to advance the promise of an immunotherapy in the treatment of multiple myeloma. Legend Biotech is seeking a Scientist, Exploratory Technology in vivo CAR-T as part of the Research and Development team based in Philadelphia, PA . Role Overview We are seeking an innovative scientist (Exploratory platform development) who possesses extensive knowledge and experience in immunology and exploratory research in cell immunotherapy to contribute to our in vivo CAR T cell platform development and to support our pipeline development. Key Responsibilities Produce research grade lentivirus to perform exploratory research. Perform lentivirus titering such as p24 ELISA and qPCR. Contribute in developing and optimizing novel cell and gene therapy platforms and in vitro cell-based immunoassays such as whole blood transduction. Setup in vitro CAR T cell culture, characterization and functional assays including PBMC culture, T cell isolation and expansion, multi-color flow cytometry, cytotoxic assays (e.g. IncuCyte), cytokine detection (e.g. MSD) and VCN analysis. Independently design and execute experiments and at the same time, work closely with the team and cross-functional teams in a highly collaborative environment. Deliver reliable and consistent data and be able to interpret scientific results and present the data within research team and cross-functional teams. Complete ELN in a timely manner with good documentation skills. Support laboratory chores as a good team player. Experience to support project from early research to clinical stage is a plus. Stay alert to industry development, keep learning and be comfortable in a fast-paced team. Requirements PhD in Immunology, Cell Biology, Molecular Biology, or a related field OR Master's degree with 3-6+ years of relevant experience. Hands-on experience in immunology, cell therapy, gene therapy, or related fields. Experience with viral vector systems (especially lentivirus) is highly preferred. Industry or postdoctoral experience is a plus. Cell biology and molecular biology techniques. Virology and viral vector systems (e.g., lentivirus). Mammalian cell culture and immune cell handling. Flow cytometry and immunoassays. Strong data analysis, organization, and problem-solving skills. The company was the brainchild of visionary scientists who recognized that antibody-based therapeutics could potentially treat disease and possessed the backgrounds in immunology and gene editing to take the leap. For two years, Legend Biotech, then known as the "Legend Project," operated in a room the size of a freight elevator, with the goal of creating a cell therapy for multiple myeloma, a hematological cancer that often relapses and can become refractory. The team produced nanobodies for single-domain antibody drugs and ultimately extended the half-life of the nanobodies with proprietary technologies. In 2015, Legend's scientists focused on research for chimeric antigen-receptor T-cells (CAR-T) targeting the BCMA protein, making the biotech one of the first companies in the world to engineer CAR-T cells for the BCMA protein. In 2016, investigator-initiated trials began in China. In 2017, data from the trials were presented at the American Society for Clinical Oncology (ASCO), which led to a partnership with biopharmaceutical company Janssen Biotech, Inc. to co-develop the anti-BCMA CAR-T, ciltacabtagene autoleucel (cilta-cel). Applications seeking approval of cilta-cel for the treatment of patients with RRMM are currently under regulatory review by several health authorities around the world.
