job summary: We are seeking a highly motivated and talented Scientist to join our In Vivo Antibody Discovery and Molecular Core located in Worcester, MA. The successful candidate will support the development of new paired antibody sequencing technologies from single B cells using droplet microfluidics. Responsibilities include experimental planning and execution, comprehensive B cells sequencing analysis and presentation, and detailed documentation to support multiple programs across our in vivo antibody discovery portfolio.
Understand the broad objectives of the project as well as their role in achieving those objectives, and modify the experimental plan when required. Set and meet experimental timelines.
Ability to work independently and as part of a team, managing multiple projects simultaneously.
Demonstrated hands-on expertise in droplet microfluidics of single B cells and next generation sequencing of IgG variable heavy and light chains is required.
Familiarity with next generation sequencing platforms (Illumina MiSeq and MiSeq i100) and knowledge of B cell genetic data analysis software and databases (FLASH, IgBlast, Usearch and IMGT) is required.
Experience in reverse transcription, RNA/DNA isolation and purification, overlap extension PCR, primer design, cDNA library generation, gel electrophoresis is required.
Protein and cell detection methods experience (ELISA and/or FACS) is preferred.

Independently design, execute and interpret experiments utilizing microfluidics to amplify heavy and light chain genes and generate DNA libraries for deep sequencing of single B cells captured in droplet emulsions.Understand the broad objectives of the project as well as their role in achieving those objectives, and modify the experimental plan when required. Molecular Techniques:
Demonstrate a high level of proficiency in conducting RNA isolation, and DNA purification, and related methods to amplify antibody heavy- and light-chain variable regions from single cells or lysates and submit DNA libraries for next generation sequencing (MiSeq/MiSeq i100).Bioinformatics Data Analysis:
Perform comprehensive in silico antibody genetic analysis and data visualization using standard and custom antibody bioinformatic tools - to guide program decisions and deliver biotherapeutic candidates.Cross-Functional Collaboration:
Collaborate with multidisciplinary teams to present/discuss experimental findings to ensure alignment with overall biotherapeutic development goals.Ability to work independently and as part of a team, managing multiple projects simultaneously.
qualifications:
Master's degree in biology, immunology, cell/molecular biology, or in a related discipline with experience in single B cell sequencing research, and 2+ (MS) years of job-related experience.Demonstrated hands-on expertise in droplet microfluidics of single B cells and next generation sequencing of IgG variable heavy and light chains is required.
Familiarity with next generation sequencing platforms (Illumina MiSeq and MiSeq i100) and knowledge of B cell genetic data analysis software and databases (FLASH, IgBlast, Usearch and IMGT) is required.
