This role is responsible for overseeing Sanger sequencing workflows, managing production schedules, ensuring data quality, maintaining sequencing instruments, and supervising laboratory technicians. The ideal candidate should have strong hands-on experience in molecular biology and Sanger sequencing, solid troubleshooting skills for sequencing data issues, and the ability to manage a fast-paced production laboratory while maintaining high standards of accuracy, efficiency, safety, and compliance. Responsibilities Supervise daily Sanger sequencing operations, including sample receiving, preparation, PCR-related processing, cycle sequencing reactions, purification, capillary electrophoresis, and data generation. Manage daily sample scheduling and production planning to ensure efficient workflow, stable throughput, and timely delivery of sequencing results. Operate and oversee routine use of mainstream Sanger sequencing instruments, such as ABI 3730XL, including daily maintenance, calibration, troubleshooting, and consumables management. Review and troubleshoot sequencing data issues, including mixed peaks, missing bases, noisy background, low signal-to-noise ratio, weak signal, short reads, and poor-quality reads. Implement and monitor QC/QA procedures throughout the sequencing workflow to ensure data accuracy and consistency. Track key production indicators, including turnaround time, repeat rate, failure rate, sample loss, reagent consumption, and overall laboratory efficiency. Train, mentor, and evaluate laboratory technicians on standard operating procedures, sequencing workflows, instrument operation, data review, and safety practices. Coordinate team schedules, assign daily tasks, and support the development of a standardized training and performance assessment system. Manage reagent and consumable inventory, purchasing requests, expiration date tracking, and cost control. Maintain instrument records, maintenance logs, calibration records, laboratory documentation, and LIMS/sample tracking information. Ensure compliance with laboratory safety requirements, 5S management, biosafety practices, hazardous waste disposal, environmental monitoring, and cross-contamination prevention. Lead troubleshooting and root-cause analysis for batch sequencing failures, customer data issues, instrument abnormalities, or laboratory incidents, and implement corrective and preventive actions. Qualifications Bachelor's degree or above in Biology, Molecular Biology, Biotechnology, Genetics, Biochemistry, or a related field. Minimum of 5 years of hands-on experience in molecular biology and/or sequencing laboratory operations. Minimum of 2 years of experience supervising a laboratory team or work group, preferably with experience leading a Sanger sequencing team of 3 or more members. Proficient in Sanger sequencing workflows and related molecular biology techniques, including DNA template preparation, PCR, sequencing reaction setup, purification, and capillary electrophoresis. Skilled in operating and maintaining Sanger sequencing instruments such as ABI 3730XL or equivalent platforms. Proficient in using sequence analysis software such as SeqMan, SnapGene, Chromas, or similar tools. Strong ability to independently analyze and troubleshoot abnormal sequencing results, including mixed peaks, missing bases, background noise, low signal, and low-quality reads.
