Purification is the highest-leverage step in the pipeline, and these four openings (two per shift) directly support the throughput and recovery targets of the operation. The Operator prepares the crude injection, loads the preparative HPLC, monitors the gradient, collects and analyzes fractions, executes pooling under the Shift Lead's direction, and runs the Dowex 1x8 acetate-form ion-exchange that yields the post-IEX solution. Key responsibilities The Operator weighs and dissolves crude peptide in the MPCR-specified solvent at the specified concentration, clarifies the injection as required, and confirms column identity, mobile-phase composition, flow rate, and detection wavelength against the MPCR before presenting the system for Shift Lead verification. During the run, the Operator monitors the chromatogram, back pressure, and detector baseline, confirms fraction-collection triggers, labels collection vessels, and manages the collector through the elution window. They support analytical RP-HPLC of fractions, record analyses in the batch record, and execute the pool under Shift Lead direction. The Operator runs the Dowex 1x8 acetate-form ion-exchange step — equilibration, loading, elution, and post-IEX consolidation — monitors residual TFA against the MPCR target, and prepares the post-IEX solution for transfer to lyophilization per the chain-of-custody SOP. The Operator cleans columns, regenerates resins, prepares mobile phases, maintains line clearance throughout the shift, and documents every cleaning and regeneration step. Required qualifications An Associate's degree in chemistry, biochemistry, or a related field is required, or a high-school diploma with substantial relevant laboratory experience, plus at least one year of hands-on HPLC or chromatography experience (preparative scale strongly preferred). The Operator must be able to follow an MPCR with full fidelity, interpret basic chromatograms, record contemporaneously per ALCOA+, and work safely with chemical PPE around pressurized fluids. Preferred qualifications Prior preparative-HPLC experience in pharmaceutical or peptide manufacturing is preferred, along with any prior cGMP experience and familiarity with Chromeleon CDS or an equivalent chromatography data system. Physical requirements and working environment On-site, shift-based; the four openings split two on Day shift and two on Evening shift. PPE throughout the shift, face shield during column packing or unpacking, standing for the full shift, lifting to fifty pounds (mobile-phase containers, column hardware), and work around running pumps, pressurized columns, and acetonitrile-TFA mobile phases.
