Across two shifts, the Operator prepares cleavage cocktails under Shift Lead supervision, executes cleavage in a fume hood, runs precipitation and ether washes, dries and weighs the crude pellet, and packages the QC sample aliquot. Key responsibilities The Operator weighs and combines cleavage-cocktail components per the MPCR, presents weighings for dual-verification, and prepares the cocktail in a functioning fume hood in acid-resistant PPE. They transfer the dried peptidyl resin to the cleavage vessel, add the cocktail per the MPCR, document the start time, and watch for any anomalous color change or temperature behavior, reporting to the Shift Lead immediately. The Operator filters the cleavage solution, washes the resin, and consolidates filtrate, then executes precipitation — dropwise addition to cold ether or MTBE with continuous stirring — and the centrifugation or filtration that collects the crude pellet, followed by repeat ether washes and drying to a free-flowing powder. The Operator weighs the dried crude, records mass and yield in the MPCR, pulls and labels the representative QC aliquot per the chain-of-custody SOP, and cleans the vessel and glassware per the validated cleaning SOP. Required qualifications An Associate's degree in chemistry, chemical technology, or a related field is required, or a high-school diploma with substantial relevant laboratory experience, plus at least one year of hands-on laboratory experience (TFA handling or chemical manufacturing strongly preferred). The Operator must follow an MPCR with full fidelity, record per ALCOA+, and work safely in acid-resistant PPE in a fume-hood environment. Preferred qualifications Prior cleavage or precipitation experience in peptide manufacturing is preferred, along with any prior cGMP experience. Physical requirements and working environment On-site, shift-based; the two openings cover the Day and Evening shifts. Acid-resistant PPE during cleavage and precipitation, standing for the full shift, lifting to fifty pounds, fume-hood work, and tolerance of ether and TFA odors at facility limits.
