Position Summary The Horlbeck Lab studies how disruption in the cellular machinery that regulates our genome and epigenetics leads to differences in how stem cells differentiate. In particular, we focus on a group of genetic disorders called “chromatinopathies,” with the ultimate goal of finding treatments for these disorders. Our work will have broader implications on the understanding of neurodevelopmental disorders and the fundamental mechanisms of gene regulation. We use cutting-edge techniques including CRISPR-based screening, single-cell RNA and chromatin accessibility sequencing, and advanced computational methods. Research will primarily be conducted in iPSC- and patient-derived neuronal models. As our lab is new, this role would involve both set-up of equipment and protocols as well as conducting independent projects. Key Responsibilities + Maintain human pluripotent stem cells and perform genetic manipulation and cell differentiation protocols. + Perform cellular analysis using techniques such as immunostaining, flow cytometry, DNA/RNA isolation, PCR, and other molecular biology methods. + Perform molecular cloning. + Conduct single-cell experiments and prepare samples for next-generation sequencing. + Operate laboratory instruments including flow cytometers and work with core facility operators for cell sorting and sequencing. + Manage inventory by monitoring, ordering, and organizing supplies. + Prepare media and stock solutions for experiments. + Ensure compliance with institutional and federal guidelines regarding health, safety, and animal use regulations. + Develop research figures and protocols while maintaining an accurate electronic lab notebook. + Mentor and instruct laboratory techniques. + Present research findings during lab meetings. + Gain additional knowledge and skills depending on interests, including computational analysis.
